Reduction in the most kinematic parameters of ram epididymal sperm in vitro by ODQ (inhibitor of nitric-oxide-sensitive guanylyl cyclase)

The present study investigated the effects of 1H-[1,2,4]Oxadiazolo[4,3-a]quinoxalin-1-one (ODQ) and ODQ + 3,3-Bis(aminoethyl)-1-hydroxy-2-oxo-1 triazene (NOC-18) on the kinematic parameters of epididymal ram sperm in vitro. After incubation of epididymal sperm samples in the presence of ODQ (1, 10, and 100 ìM) alone or with NOC-18 (0.0005 ìM), motion parameters were evaluated at 45 and 90 min by computer-aided sperm analysis. There was significant (P<0.05) decrease in the fast progressive motility, curvilinear velocity, straight line velocity, average path velocity, straightness, mean angular displacement, amplitude of lateral head displacement, and linearity of sperm in ODQ-treated or (ODQ + NOC-18)- treated groups compared to their controls. On the other hand, there was significant (P<0.05) increasing of immotility and beat cross frequency compared to control. It is concluded that physiological function of nitric oxide on motion parameters of epididymal sperm is mediated via guanylyl cyclase/cGMP pathway in ram.
